The Des Moines Register recognised him as the top boys prep athlete in the state. As a true freshman, he went on to start seven games at right tackle and became the first player in the Kirk Ferentz era to start at offensive tackle.

After his sophomore season, he broke Brandon Scherff's record for the Hawkeyes hang clean, setting a new record at 450 pounds. Wirfs declared that he would forego his senior season and enter the 2020 NFL Draught after his junior year, during which he was named to the First Team All-Big Ten and the league lineman of the year. Tristan was suspended for an OWI charge later in his college career after it was discovered that he had driven drunk. However, he went on to be named conference lineman of the year and made the First Team All-Big Ten. After being selected by the Tampa Bay Buccaneers with the 13th overall choice in the first round of the 2020 NFL Draught, Tristan helped the team win Super Bowl LV.
