She is the daughter of parents, an American mother from Virginia, and a Colombian father from Pereira. On July 17, 1994, he was born in Alexandria, Virginia with birth name Karly-Marina Loaiza. During the late 1980s, Uchis' parents met. She resided with her mother and elder siblings throughout the academic year. Kail learned to play the piano and saxophone, while in high school. 

She frequently missed class to work on experimental short films in the photo lab. She made mixtape cover art as a result of her love of photography. Uchis made her debut EP Por Vida available for free download in February 2015 on her own website and afterward on iTunes. The song "She's My Collar" and the extra tracks "Ticker Tape" and "She's My Collar (Kali Uchis Spanish Special)" are from Gorillaz's Humanz, their fifth studio album, which was released in April 2017. The second single from the album, "Nuestro Planeta," featuring Reykon, was later made available by Uchis on August 25, 2017. On October 30, 2018, the fourth and final single from the album, "Just a Stranger" with Steve Lacy, was made available.

American rapper Mac Miller and Uchis worked together on the song "Time" in June 2019. It was Miller's first official postmortem release since his passing on September 7, 2018, and it was written by Uchis. Uchis released "Solita" in December 2019, which was initially intended to serve as the lead single of her upcoming second studio album but was subsequently taken off the final tracklist. Uchis revealed on the red carpet at the Latin American Music Awards on April 21, 2022, that she had finished recording her third and fourth studio albums, one in Spanish and one in English. On her social media, she posted a teaser of the house-influenced song "No Hay Ley," which was then made available as a single on September 2, 2022.
